Solubility Study
Solubility measurement data is an integral component during HTS screening, during formulation and during regulatory preparation and submission - solubility decreases as molecular weight increases and molecular diversity increases with higher molecular weight. At Robertson Microlit, solubility measurements are performed at specific pH values using a modified ‘shake-flask’ method and solutions quantified by UV/ VIS spectrophotometry. The analysis is carried out in gastric fluid @ pH 1.2, intestinal fluid @ pH 7.4 and buffer @ pH 7.0. Additional media are available to meet specific requirements. At least 15.0 mg. of sample and an absorbing chromophore is required for measurement. Our solubility screening program includes measurements made with several solvents at one specific solvent and a fixed experimental time. All data is stored on disk with reports including:
-
solubility expressed in mg/mL for each medium
-
reference spectrum in methanol
-
maxima, concentration, weight, absorbance
